Summary, Etc.A series of experiments were conducted to develop the necessary protocol for shoot culture of different banana cultivars. The optimum conditions for producing plantlet through in vitro system using corm as initial material was determined. The necessary protocol for shoot culture of banana using corm as initial material was formulated. Corms were surface-sterilized in pure commercial bleach (5.25% sodium hypochlorite). Freshly harvested corms required 30 minutes sterilization while stored corms required 45 minutes. Apical shoot was cultured aseptically on a solidified nutrient medium. The composition of the culture medium found suitable for the multiplication of banana shoots were derived. Transfer of most cultivars to fresh medium required six to ten weeks after initial inoculation. Where blackening around the corm was severe, frequent transfer (every two weeks) alleviated the problem. Individual shoots from in vitro culture were separated and subcultured into fresh medium. Multiplication was done by transferring plants less than 2 cm or by sectioning the shoot of larger plants into two or four to encourage proliferation. Shoots of some cultivars such as Latundan and Cardaba were increased by sectioning. Production of shoots from explanted corm sections and subcultured shoots of different bananas and plantains were elicited by culture medium. In some, adding naphthalene acetic acid at 1 mg/l and supplementing benzyl adenine at lower concentration (1 mg/l) to the medium enhanced the number of shoots produced per culture. When sufficient shoots were formed, individual shoots were excised and transferred to a rooting medium. The time of planting, sterilization, potting media and fertilization of rooted shoots were determined
