A colorimetric procedure was developed for determining the numbers of Rhizobium japonicum (soybean rhizobia) in legume inocula. The procedure is similar in principle with enzyme-linked immunosorbent assay (ELISA) which employs alkaline phosphatase, anti-rhizobia IgG complex. Some technical parameters that were defined and tested were purification of rabbit-derived antibodies against Rhizobium japonicum; conjugation of purified antibodies with an alkaline phosphatase enzyme; heating of bacterial suspensions to destroy non-specific antigens; labelling bacteria with the antibody-enzyme complex, removing non-adsorbed enzyme prior to chemical analysis and defining conditions of the enzyme assay used to determine cell numbers.
