Summary, Etc.Biochemical and genetic studies were conducted for four nodule enzymes of mungbean (Vigna radiata L. Wilczek) and yardlong bean (V. unguiculata L. Walp.) namely: glutamine synthetase (GS), glutamate synthase (GOGAT), glutamate dehydrogenasse (GDH) and phosphoenolpyruvate carboxylase (PEPC). Kinetic studies revealed close similarities in Km values, pH and temperature optima and temperature stabilities of the four nodule enzymes from mungbean compared with the same nodule enzymes from yardlong bean. Results of the genetic studies showed significant variation in the activity levels of GS and PEPC among the various mungbean and yardlong bean genotypes evaluated. High and low nodule GS and PEPC genotypes were identified from the 12 mungbean and 12 yardlong bean samples tested. Significant variation were also detected in other nitrogen fixation traits tested. Multiple regression analysis was conducted between nodule enzyme activities and shoot and nodule traits, and the amount of nitrogen fixed in mungbean genotypes. Only shoot dry weight and nodule number gave significant tests for general combining ability (GCA) and specific combining ability (SCA) effects for nodule GS and PEPC activities in both species. Significant GCA effects were likewise obtained for GOGAT and GDH activities in mungbean and yardlong bean. Mungbean and yardlong bean genotypes which are best combiners for nodule enzyme activities were identified. The implications of the results in nitrogen metabolism, speciation and breeding fixation were discussed
