Six primer pairs were designed from the biphenyl (bph) operon of Pseudomonas pseudoalcaligenes KF707 to detect dehydrogenase genes from ciliates using polymerase chain reaction (PCR) technique. Optimum PCR conditions were obtained empiricallyand had much lower annealing temperatures than those suggested by the OLIGO 4.0-s primer design program. Four (4) PCR products were obtained from 3 out of6 species of ciliates used in the study. Two (2) products, AIL and Al Tm of primer bphAI are fromParamecium aurelia complex (L) and from Tetrahymena sp. (Tm), respectively. BTI and CTI, both from Tetrahymena thermophila (TI) are products of primers bphB and bphC, respectively. Analysis of the sequenced PCR products using BLASTN and F ASTXY programs strongly suggests that AlL, Al Tm and BTI sequences are fragments of dehydrogenase genes. These are not only potential PCB-degrading genes but may also be involved in the degradation pathway of other xenobiotics. Among the 3 sequences, the BTI sequence had perfect annealing sites for both the sense and anti-sense primers. The CTI sequence differs from the other products as shown by BLASTN and F ASTXY search in that it is 100% homologous to dynein heavy chain of 7: thermophila.
