aCloning and transformation systems were developed in the basidiomycete clitopilus passeckerianus NRRL 3100. Five strains of C. passeckerianus were used, namely the wild type strain (Cpwt), HP76, LP1, LP2 and LP5. Genomic DNA was isolated from the five strains and used as template for polymerase chain reaction (PCT. The primers used were constructed based on GENBANK sequences of the geranylgeranyltransferase (GGT) gene, one of the genes involved in the biosynthetic pathway of pleuromutilin. A 200-bp product was obtained from primer pairs GGT2F/GGT2R and GGT3F/GGT3R using Cpwt as template. Degenearate primers based on GGT were also constructed. From the degenerate primers, five bands were obtained from Cpwt, eight bands for LP5, while no PCR products were obtained from HP76, LP1 and LP2.0
