Leaf explants of Jatropha curcas Linn, were collected from 2 plants and cultured in full strength MS medium supplemented with factorial combinations of the hormones Naphthaleneacetic acid (NAA) and 6-Furfurylaminopurine (Kin) (0 uM, 5 uM, 10 uM, 30 uM and 50 uM). The earliness of callus induction (average number of days) was monitored, and for Plant 1 five combinations of the hormones were able to produce callus at a significantly earlier time (5 uM NAA + 10 uM Kin, 10 uM NAA + 30 uM Kin, 10 uM NAA + 5 uM Kin, 50 uM NAA + 10 uM Kin and 30 uM NAA + 30 uM Kin), producing callus after 5 to 7 days of culture. For Plant 2, media supplemented with 50 uM NAA + 10 uM Kin was first to produce callus after an average of 6 days. The average fresh weight (FW) callus produced by Plants 1 and 2 was highest for treatments supplemented with 10 uM NAA + 5 uM Kin, and 10 joM NAA + 10 uM Kin. Furthermore, for the 2 plants the absence of either of the hormones caused no callus production, late induction or low amounts of callus. Also for both plants, high amounts of either or both hormones (50 uM and, to a lesser effect, 30 uM) can also cause negative effects on the induction and growth of callus. For both plants, dry weight (DW) constituted only a small percentage of the FW callus. Analysis by HPLC of the callus of J. curcas (both plants) was able to show a peak at elution time of 3 to 4 minutes, showing that diterpenoids were produced by the callus. Variation in the relative amounts of diterpenoids produced was observed among the different treatments, but there was no obvious trend showing any relationship between the treatments and the relative amounts produced by the calli.
